METHODS:HumanplateletaggregationsinducedbyA DP,adrenaline,andcolagenweremeasuredturbidi metricaly.Theaggregationcurvewasrecordedona plateletaggregometerandthemaximalaggregatio nrate(ARmax),efectivedeaggregationratein5mi n(DR5min)andlagtime(LT)wereautocalculatedby thebuiltinmicrocomputer;PF4liberationfromh umanplateletsstimulatedbyADPandcolagenwasde terminedbyrecordingtheheparinthrombinclotin gtime(HTCT).Thrombosiswastestedbyweighingth ewetanddrythrombiformedinasiliconizedrevolv ingring.RESULTS:AGVPSinhibitedinvitrotheARm axwithIC50of133(95%confidencelimits:109-1 63,ADPinduced),166(154-179,adrenaline induced),and42g·L-1(06-29,colageninduced ).TheDR5min(onADPinducedaggregation)andLT( colageninduced)werealsoincreasedaswel.Mean while,AGVPS063-250g·L-1prolongedHTCTonADP andcolagenstimulatedPF4liberation.At034- 139g·L-1,AGVPSreducedthewetanddryweightoft hrombiformedinvitro.CONCLUSION:AGVPSinhibit shumanplateletaggregation,liberation,andthr ombosisinvitro,suggestingitspossibleantithr omboticactioninman.Thetotalsaponinsextracte dfromAcanthopanaxgracilistylusvarpubescens (pampanin)Li(AGVPS),showedprotectiveefectso nmyocardialischemia〔1〕,hypercholesterolemia 〔2〕,andbeneficialefectsagainstarhythmiasind ucedbychloroform,BaCl2,isoprenaline,andouab aine〔3〕aswelasbyischemiaandreperfusion〔4〕.T heplateletplaysanimportantroleinthedevelopm entofcoronaryheartdiseasesandarhythmias〔5-6 〕.Ourpresentworkwasundertakentostudytheefec tsofAGVPSonhumanplateletaggregation,Platele tfactor4(PF4)liberation,andthrombosisinvitr o.MATERIALSANDMETHODSDrugsandreagentsAGVPS, ayelowishcrystalpowder,wasextractedfromtheC hineseherbAcanthopanaxgracilistylusvarpubes censwithclasicalmethod〔7〕anddeterminedinGua ngzhouInstituteofMedicalandPharmaceuticalIn dustry.ThesaponincontentofAGVPSwas289%dete rminedwithphotocolorimetricmethod.Tenspotso fginsenosideswereshownonthesilicagelthinla yerplate,fourofthemhavingthesameRfvaluesasR b1,Rd,Re,andRgcomponentsofginsenosides(Labo ratorydataofHunanMedicalUniversity).Itwasdi ssolvedinnormalsalineandthepreparationswere freshlymadebeforeeachuse.ADPwaspurchasedfro mFlukaChemicalCo,Switzerland,lot№263138387 .Adrenaline(lot№910118)andthrombin(lot№9208 12)wereproducedbyGuangzhouMinxinandZhuhaiBi ochemicalPharmaceuticFactories,respectively .Theyweredissolvedin02mol·L-1phosphatebufe rsolution(PBS,pH72)andstockedat-20℃.Colage nwasmadefromrabbittendon〔8〕andstoredat4℃for <4d.PlateletaggregationtestVenousbloodwasob tainedbyveinpuncturefromhealthycolegestuden tsofeithersex,aged21±2a,whohadtakenneitherm edicinenorgreasyfoodforatleast2wk.Thebloods ampleswereanticoagulatedwith34%sodiumcitra te(9∶1,vol/vol)andcentrifugedat190×gfor5min togetplateletrichplasma(PRP).Theprecipitat ewascentrifugedat1800×gfor10mintoobtainthep lateletpoorplasma(PPP).Theplateletcountine achPRPwasadjustedto25×1011·L-1bydilutionwi thPPPfromthesamesample.PRP02mLwasplacedina curvetteandstiredwithAGVPSorcontrolsolution25μLat37℃for5min,thenaggregatingagent25μLwasadded(finalconcentration:ADP47μmol·L-1,adrenaline62μmol·L-1andcolagen40mg·L-1).Thep
