±20)g,provided by the center oflaboratory anim alscience of Guangdong (the quality exequatur num ber of No.2002A024),were adopted,and totally 24ratswere random ly cho-sen from them and divided into preventive group (12rats)and ther-apeutic group (12rats)ofsham operation (only opened the rats’abdom en butnotrem oved their ovary), and the left48rats wererandom ly divided into 4groups after replicating OP m odel by o-variectom y (which opened the rats’abdom en and rem oved theiro-vary): OP m odelcontrolgroup ofprevention, OP m odelcontrolgroup oftreatm ent,OP m odelgroup ofprevention with yigu capsuleand OP m odelgroup oftreatm entwith yigu capsule,with 12rats ineach group.M edicine:Yigu capsule was m ade up by epim edium ,fruitoflycium barbarum l,radix angelica sinensis,roots ofachyran-thes bidentata blum e etc.,the decoct-free granule ofthe drugs wereprovided by Shenzhen Sanjiu M edical&Pharm aceuticalCo.,Ltd,the aqueous solution was m ade to include crude drug 1.0g/m L.M ethodsThe preventive groups ofyigu capsule were adm inistered by gastricinfusion 3days afterovariectom y,3m L NS was given to preven-tive group of sham operation and OP m odel control group and 3m L aqueous solution ofyigu capsule was given to preventive groupwith yigu capsule, 1tim e a day for 24weeks; the therapeuticgroups ofyigu capsule were adm inistered by gastric infusion in the13th week after ovariectom y, 3m L NS was given to therapeuticgroup of sham operation and OP m odel control group and 3m Laqueous solution of yigu capsule was given to therapeutic groupwith yigu capsule,1tim e a day for12weeks.M aderatstodeath 24weeksafterovariectom y,separated thighbone,slices were m ade by SP m ethod ofim m unohistochem istry,the dis-tributed situation ofBM P-2wasobserved by optic m icroscope,andpalebrown particleswould appearin thecytoplasm ofpositive dyeingcells.LEICA Qwin pictorialanalyticalsoftwaresystem wasadopted tom ake a half-quantitative grey scan, randomly chose 10eyeshotineach slice and determ ined the M G of the com parative content ofBM P-2positive reaction cells,in which the M G ofunitarea wasre-garded asparam eterofexpressingquantity.Statisticalanalysis:SPSS 12.0wasused forstatisticalanalyze by thefirstauthor,ANOVA wasadopted forthedatum analyze,the data wasexpressed byM ean ±SD,two-sided α=0.05wasused fortestcriteria.R ESU LTSQ uantitative analysis ofthe experim entalanim alsTotally 72rats were adopted in the experim entand allentered intothe analysis ofresults,no one dropped off.StatisticalinferenceResults ofthe M G ofBM P-2positive reaction cells in bone tissueIn the 24th week afterovariectom y,the M G ofBM P-2positive reac-tion cells in bone tissue of preventive OP m odel control group,preventive group with yigu capsule and preventive group of shamoperation were 81.84±10.01,124.46±6.55,130.15±10.09,thatofpreventive OP m odelcontrolgroup was obviously lowerthan thatofpreventive group ofsham operation (P <0.01),and thatofpreven-tive group with yigu capsule was obviously higherthan thatofpre-ventive OP m odelcontrolgroup (P <0.01)and closed to thatofpre-ventive group ofsham operation (P >0.05).The M G ofBM P-2posi-tive reaction cells in bone tissue of therapeutic OP m odel controlgroup,therapeutic group with yigu capsule and therapeutic group ofsham operation were 77.97±10.35,110.32±8.69,131.79±12.10,thatof therapeutic OP m odel control group was obviously lower thanthatoftherapeutic grou
